چکیده
Lithospermum erythrorhizon (L. erythrorhizon) root is used in traditional medicine for its anti-inflammatory, antibacterial, and antioxidant properties. However, no studies have examined impact on allergic rhinitis (AR). Here, we explored the protective effects of L. immunoglobulin E (IgE)-stimulated RBL-2H3 cells an ovalbumin (OVA)-induced AR mouse model. In latter, nasal mucosal inflammation, allergen-specific cytokine production, histological changes to mucosa. model, oral administration ethanol extract (LE) led a marked reduction rubbing sneeze frequency, significant decrease serum OVA-specific IgE IgG1 levels, increase IgG2a/IgG1 ratio. LE also reduced expression interleukin (IL)-4, IL-5, IL-13 lavage fluid (NALF), suppressed inflammatory cell infiltration epithelial degradation tissues. IgE-stimulated cells, release degranulation markers such as β-hexosaminidase histamine. Based these findings, suggest that may ameliorate OVA-induced by regulating mast cell-mediated responses.

IgE alone-induced actin assembly modifies calcium signaling and degranulation in RBL-2H3 mast cells.
In the mast cell signaling pathways, the binding of immunoglobulin E (IgE) to FcepsilonRI, its high-affinity receptor, is generally thought to be a passive step. In this study, we examined the effect of IgE alone, that is, without antigen stimulation, on the degranulation in mast cells. Monomeric IgE (500-5,000 ng/ml) alone increased cytosolic Ca2+ level ([Ca2+]i) and induced degranulation in r...
متن کاملEstrogen mimetic 4-tert-octylphenol enhances IgE-mediated degranulation of RBL-2H3 mast cells.
Allergic diseases such as asthma have been on the rise in recent decades. Environmental or occupational exposure to estrogenic synthetic chemicals is suspected to be a contributing factor, and previous experimental studies indicated that estradiol and some xenoestrogens increase allergic signaling responses, such as degranulation, in immune cells. In the current study, data showed that the estr...
متن کاملInhibitory Effect of Methyleugenol on IgE-Mediated Allergic Inflammation in RBL-2H3 Cells
Allergic diseases, such as asthma and allergic rhinitis, are common. Therefore, the discovery of therapeutic drugs for these conditions is essential. Methyleugenol (ME) is a natural compound with antiallergic, antianaphylactic, antinociceptive, and anti-inflammatory effects. This study examined the antiallergic effect of ME on IgE-mediated inflammatory responses and its antiallergy mechanism in...
متن کاملInhibitory effect of ellagitannin metabolites on IgE-mediated allergic responses in RBL-2H3 cells.
The effects of four ellagitannin metabolites (M1-M4) and ellagic acid on immunoglobulin E-mediated allergic responses in rat basophilic leukemia-2H3 cells were investigated. M1-M4 inhibited the antigen-induced degranulation and secretion of interleukin-4 and tumor necrosis factor-α, but ellagic acid only slightly did so under the same experimental conditions. M1 inhibited the activation of the ...
متن کاملInhibitory Effect of Loranthus parasiticus on IgE-Mediated Allergic Responses in RBL-2H3 Cells
The mistletoe Loranthus parasiticus has been used as a compound for traditional medicine in Northeast Asia for a long time and is known to possess neuroprotective action. Nonetheless, the effect of Loranthus parasiticus on allergic responses remains unknown. In the present study, we evaluated whether the water extract of Loranthus parasiticus (LPE) could inhibit IgE-mediated allergic responses ...
